About this project
The objective of the PhD project is to develop colon-derived organoids that recapitulate the cystinosis phenotype of variable CTNS mutants using CRISPR/Cas9 technology on healthy colon organoid lines. These organoids will be thoroughly characterized for their cystinosis phenotype, compared with cystinosis tubuloids and cultured as microtissue in in-house designed microfluidic devices suitable for drug screening in a medium throughput fashion using 3D fluoresecent imaging. Drugs will be tested for their ability to restore the cystinosis phenotype in organoids and tubuloids.
